However\, Pleiotropy o ccurs when a genetic variant influence the response though multiple phenot ypes of interest\, and therefore renders it an invalid instrumental variab le in MR studies. We propose a novel Smoothed Constrained Instrumental Var iable (CIV_smooth) method to construct valid instrumental variables while correcting for pleiotropic phenotypes. In a series of simulation studies\, we show that our method leads to causal effect estimation with reduced bi as compared with popular methods. Finally\, we analyzed Alzheimer’s diseas e data from Alzheimer’s disease (AD) Neuroimaging Initiative (ADNI) study\ , and identified significant causal effect of Amyloid beta on AD progressi on using CIV_smooth method. However\, in view of the limitations of this s tudy\, we understand further work are still needed to make substantive cau sal statements.\n DTSTART:20180322T190000Z DTEND:20180322T200000Z LOCATION:Room PK-5115 \, CA\, UQAM\, Pavillon Kennedy SUMMARY:Lai Jiang\, 9IÖÆ×÷³§Ãâ·Ñ URL:/mathstat/channels/event/lai-jiang-mcgill-universi ty-285893 END:VEVENT END:VCALENDAR
