However\, the details\nof this view a re largely unknown. To explore in this direction\, we\ndeveloped the Chrom atin Interaction Analysis using Paired-End-Tag\nsequencing (ChIA-PET) stra tegy for de novo detection of genome-wide\nchromatin interactions\, and de monstrated this approach through\ncomprehensive mapping of chromatin inter actions involved in\ntranscription regulations mediated by estrogen recept or α (ERα) in\na human genome. In order to map all chromatin loops that ar e\nimportant to chromatin architecture and gene transcription\nregulation\ , we have applied the ChIA-PET strategy to general\nprotein factors such a s RNA polymerase II (RNAPII) involved in all\ntranscription activities as analysis targets in a number of human\ncells. Our recent results reveal th at large numbers of RNAPII genes\nare organized into operon-like configura tion through long-range\nchromatin interactions for coordinated transcript ion regulation.\nFurthermore\, through CTCF ChIA-PET analysis we have disc overed the\nprimary role of CTCF is to confer genome-wide 3D framework of \nchromatin loops for nuclear functions such as transcriptional\nactivatio n and repression.  Further analyses of chromatin\ninteractome\, transcript ome\, and histone modification maps will\nprovide deep insights to advance our understanding of human genome\nbiology.\n \nMandatory for Biochemistr y Graduate Students\n DTSTART:20101101T160000Z DTEND:20101101T160000Z LOCATION:McIntyre Medical Building\, CA\, QC\, Montreal\, H3G 1Y6\, 3655 pr omenade Sir William Osler SUMMARY:Biochemistry and Systems Biology Seminar - Dr. Yijun Ruan (Beatty L ecture) URL:/channels/event/biochemistry-and-systems-biology-s eminar-dr-yijun-ruan-beatty-lecture-168265 END:VEVENT END:VCALENDAR
